I’ve got a 2000 model 3.4 v6, the problem I have is when I switch over to my sub tank, within 40-50 kms the tank will be empty but all the fuel has been transferred to the main tank. Could this be due to a faulty fuel pressure regulator?

It won't be the fuel pressure regulator as there is only one fitted.
Both fuel tanks have internal high pressure fuel pumps so when you switch tanks the single fuel return line has a valve which diverts the majority of the fuel to the tank selected.
It sounds like the valve is not functioning properly and the excess fuel returning is still going back to the main tank. The valve is solenoid operated and is under the passenger side just forward of the sub tank.
