Nope, you can engage it whilst moving.
You need to be stopped though to go from HL (centre diff locked, high range) to L (centre diff locked, low range)
Off memory to go back from L to HL, you need to be stopped and in neutral<Rob> '12 Prado GX - https://www.pradopoint.com.au/forum/general/my-rig-build-up/32702-i-m-back-with-a-2012-150-gx-toorak-tractor

I found that going from HH to HL is fine while moving however I needed to be going under 30 odd km/hr to get it back into HH otherwise the centre diff light didn't go away. Definately stop before engging/disengaging L.2001 Prado GXL 3.4L Petrol. Genuine Toyota Bull Bar, Tow Bar and Side Steps, Electric Trailer Brake, Spotties, Rhino Roof Racks. Nothing too fancy....yet.

Engaging the diff lock while moving is fine as long as you aren't spinning wheels as this will (s)mash the teeth on the diff lock clutch.Declan: 1997 GXL 4.5 Auto 80 series on LPG, ARB bullbar, 2" OME lift, 32" MTZ, GME UHF, rear ARB locker, Kaymar spare wheel carrier
PLANNED: 3 or 4" suspension lift, 2" body lift (maybe), winch, 35" MTZ + roadies, front locker, sliders, cylinder head rebuilt for LPG, Garret Turbocharger
